From Natural samples to Microalgal Cultures (1-5 Oct. 2012)

The BEA (Banco Espa├▒olde Algas) offers a 5-day intensive 'hands-on' training course on techniques for isolation, purification and growth of microalgae from natural samples to clonal, axenic culture.

We offer an intensive five-day training course covering basic and advanced techniques for isolating, purifying and growing microalgae. The course is designed for academic graduate students, faculty members and algal biotec personnel.

Isolation of microalgae begins with sampling. The course starts with an excursion to diverse microalgal habitats on Gran Canaria to demonstrate different sampling strategies and methods for characterization of a habitat. Direct isolations and enrichment techniques will be performed. Direct isolation includes single-cell isolation by micropipette (microcapillary and dip and drag technique), streak and spray plating, and single cell flow cytometric sorting. Enrichment techniques include serial dilution, phototaxis, and use of diverse culture media. Methods to obtain axenic cultures (e.g. ultrasonication/spray plating, single cell flow cytometric sorting) will be provided. Preparation and sterilization of various culture media will be described as well as methods to test for sterility of cultures. Methods for maintenance of microalgal cultures from Phycomat to cryopreservation will be demonstrated.

Other topics will include a discussion of factors influencing microalgal growth from stock cultures to pilot plant.

Participants are encouraged to bring their own samples for isolation and/or purification.

The participating faculty combined has more than 100 years of experience in research with microalgae including operation of large algal culture collections and isolation of thousands of strains
